Effect of Bupivacaine on the Endothelium-Dependent Relaxation in Isolated Rat Aorta

Abstract
|
|
|
This study was designed to evaluate the effect of bupivacaine on the acetylcholine-induced endothelium-dependent relaxation of isolated rat aorta in the presence of high potassium chloride and LNMMA.
Bupivacaine(100 M) inhibited the endothelium-dependent relaxation induced by acetylcholine but had no effect on the endothelium-independent relaxation induced by nitroprusside. Bupivacaine inhibited the contraction induced by calcium chloride
addition
to the clacium-free and high potassium-containing modified Krebs solution. Bupivacaine inhibited the endothelium-dependent relaxation in the high calcium-containing modified Krebs solution, Inhibiton of the endothelium-dependent relaxation by
bupivacaine was disappeared in the high potassium-containing modified Krebs solution but not in the LNMMA-treated cases. These results suggest that inhibitory effect of bupivacaine on the endothelium-dependent relaxation may be due to suppression
of
EDHF action.
